Biochemistry Laboratory II, CHE 453L

Monday and Wednesday 1:00-3:50 PM, NSM C-339

Instructor: Dr. Sturm, nsturm@csudh.edu, and Kevin Nguyen, knugyen@csudh.edu

Office Hours: TBA.

Prerequisites: (a) CHE 451L and (b) concurrent enrollment in CHE 452

Laboratory Manual: Print, http://www2.csudh.edu/nsturm/, place in a binder and bring to the laboratory:

Experimental Protocol Glucose-1-Phosphate

Experimental Protocol Glucose-1-Phosphate Characterization

Experimental Protocol Wheat Germ Acid Phosphatase Purification

Experimental Protocol Wheat Germ Acid Phosphatase Kinetics

Laboratory Reports:
One lab report for each experiment is required and will be graded for content, discussion of theory and instrument components, quality of data, correctness of calculations, closeness of experimental results to literature values, justifications for poor results, discussion of error, and as outlined in the scoring rubric.

Potato Scoring Rubric (COVID-19 Modified)

Wheat Germ Scoring Rubric (COVID-19 Modified)

Written Quiz: Will cover material from both of the laboratories (100 pts). Types of material to emphasize: What reaction does wheat germ acid phosphatase catalyze? What does 7-min hydrolysis followed by the Fiske-Subbarow assay tell us? What do the results of the Biuret assay tell you in the wheat germ lab? vs the results of the phosphatase assay? What is phenyl mercuric nitrate used for in the potato lab?

Writing Assignment: A paper word-processed, double-spaced, 250 words or less, addressing the following tasks is due on Wednesday, May 6th, 2020 (50 pts).

Unexpected challenges in our lives sometimes bring out the best in us. For example, we act heroically or courageously in emergencies, we are charitable when disasters affect our friends, or we are unselfish and patient when misfortune strikes our friends or relatives. Pick an instance in your own life when such a challenge stimulated in you a response that pleased you. Describe the situation, explain how you responded, and tell what you learned about yourself from the experience.

This paper will be judged on rhetoric, how effectively you address the writing tasks and syntax, grammar.

Grading Criteria: Letter grades are based on the following point distribution. Grades are criterion-referenced not norm-referenced; each student has an equal opportunity meet the criteria charted below:

Lab Reports 200 pts
Quiz 100 pts
Writing Assignment 50 pts
Total 350 pts
